# Alpha-MSH Explained: Function, Research & the KPV Link

**By TelosRX Editorial Team** · 2026-10-30

**Alpha-MSH is a 13-amino-acid hormone your body makes from a precursor called POMC. It shapes skin pigment, appetite, inflammation and sexual function. [KPV](https://telosrx.com/pages/kpv), a popular research peptide, is its last three amino acids.**

Most people meet alpha-MSH backward. They hear about KPV, Melanotan II or PT-141 first. Then they learn all three trace back to one small hormone.

That hormone is alpha-melanocyte-stimulating hormone, or alpha-MSH. It's worth understanding, because it explains why these very different peptides behave the way they do.

## What Is Alpha-MSH?

Alpha-MSH (α-MSH) is a peptide hormone and neuropeptide (a messenger used by nerve cells). It belongs to the melanocortin family. Its name comes from its first-discovered job: telling pigment cells to darken skin.

The basics:

- **Length:** 13 amino acids.
- **Sequence:** Ser-Tyr-Ser-Met-Glu-His-Phe-Arg-Trp-Gly-Lys-Pro-Val.
- **Made in:** the pituitary gland, hypothalamus, skin, immune cells and gut.
- **Modified ends:** one end is acetylated and the other amidated, which helps stability.
- **Family:** melanocortins, which also include ACTH, beta-MSH and gamma-MSH.

Researchers have long described it as a natural brake on inflammation. A [2014 review on NIH's PubMed Central](https://pmc.ncbi.nlm.nih.gov/articles/PMC4130143/) calls it an emerging anti-inflammatory and antimicrobial peptide.

## Where Alpha-MSH Comes From: POMC

Alpha-MSH starts life inside a larger protein called proopiomelanocortin, or POMC. Think of POMC as a long baguette that enzymes slice into several hormones.

1. An enzyme called PC1 cuts POMC into ACTH and other fragments.
2. A second enzyme, PC2, trims ACTH down.
3. One product of that trim is alpha-MSH.
4. Final tweaks to each end make the active hormone.

Fun fact: ACTH, the stress hormone that drives cortisol, contains the full alpha-MSH sequence in its first 13 amino acids. Same baguette, different slice.

## How Alpha-MSH Works: The Melanocortin Receptors

Alpha-MSH acts by binding melanocortin receptors, labeled MC1R through MC5R. Each sits in different tissues, so each controls a different job.

Receptor

Main location

Linked functions

Alpha-MSH binds?

MC1R

Skin pigment cells, immune cells

Pigmentation, inflammation signaling

Yes, strongly

MC2R

Adrenal glands

Cortisol release

No (ACTH only)

MC3R

Brain, immune cells

Energy balance, inflammation

Yes

MC4R

Brain (hypothalamus)

Appetite, energy use, sexual function

This receptor map is the key to the whole family. A drug that hits MC4R behaves very differently from one that hits MC1R.

## What Does Alpha-MSH Do? Four Main Roles

### 1\. Skin pigmentation

At MC1R, alpha-MSH tells melanocytes (pigment cells) to make more eumelanin, the darker pigment. This is part of how skin tans after sun exposure. Variants in the MC1R gene are linked to red hair and fair skin.

### 2\. Appetite and energy balance

In the hypothalamus, POMC neurons release alpha-MSH onto MC4R. That signal lowers food intake and raises energy use. Leptin, the fat-tissue hormone, switches these neurons on. Rare MC4R gene variants are among the most common single-gene causes of severe early-onset obesity.

### 3\. Inflammation and immune balance

Alpha-MSH dials down inflammatory signaling. A [1998 study in the Journal of Immunology](https://pubmed.ncbi.nlm.nih.gov/9743348/) found it blocked activation of NF-κB, a master switch for inflammation, in cell experiments. It also lowers several inflammatory cytokines (immune signaling proteins) in lab and animal models.

### 4\. Sexual function

Brain melanocortin signaling also influences sexual arousal. This link was noticed when early tanning compounds produced unexpected arousal effects in research volunteers. It later led to an FDA-approved drug, covered below.

Researchers have also studied alpha-MSH for antimicrobial activity, fever control and nerve protection. Most of that work is preclinical.

## Alpha-MSH and KPV: The Tripeptide Connection

Here's where alpha-MSH meets one of the most-searched research peptides. KPV is lysine-proline-valine. Those are positions 11, 12 and 13 of alpha-MSH, its final three amino acids.

Scientists isolated that tail because the anti-inflammatory activity seemed to live partly there. The pigment-darkening part of alpha-MSH sits elsewhere in the chain. So KPV is studied as a way to keep the anti-inflammatory signal without the tanning.

- **Gut research:** a [2008 study in Gastroenterology](https://pubmed.ncbi.nlm.nih.gov/18061177/) found KPV entered intestinal cells through a transporter called PepT1 and reduced inflammation in mouse colitis models.
- **Skin research:** early lab studies explore KPV in skin inflammation and wound models.
- **Human data:** controlled human trials are lacking.

## Alpha-MSH Analogs: The Family Tree

Natural alpha-MSH breaks down within minutes in the blood. So chemists built longer-lasting analogs (lab-made lookalikes). Some became FDA-approved drugs. Others remain research compounds.

## Alpha-MSH vs KPV vs Melanotan II

These three get mixed up constantly online. Here's the short version:

- **Alpha-MSH:** the natural hormone. Broad effects across pigment, appetite, inflammation and arousal.
- **Melanotan II:** a synthetic, longer-lasting cyclic analog. Hits several receptors, so it brings tanning and arousal together, along with side effects.
- **KPV:** just the last three amino acids. Studied for inflammation, without the pigment effect.

Same family, very different personalities. That's why "it's all alpha-MSH" is a misleading shortcut.

## Can You Measure Alpha-MSH Levels?

Some specialty labs offer a blood test for alpha-MSH. It's rarely part of routine care. A few practitioners order it when exploring chronic inflammatory symptoms, but its clinical value is debated.

Low or high results are hard to interpret alone. Levels vary with time of day, stress and medications. If you've seen this test online, discuss it with a provider before ordering.

## Safety: What Is Known

Natural alpha-MSH is something your body makes all day. Lab-made analogs are a different story, and side effects depend on which receptors they hit.

- **MC1R activation:** skin darkening and changes in moles or freckles.
- **MC4R activation:** nausea, flushing, headache and temporary blood pressure rises have been reported with bremelanotide.
- **Unregulated products:** "research use only" peptides may be mislabeled or contaminated.

Anyone with a history of melanoma or unusual moles should be especially cautious with pigment-active compounds. Your provider will review your full health history first.

## The Bottom Line on Alpha-MSH

Alpha-MSH is a small hormone with a big job list: pigment, appetite, inflammation and arousal. Its receptor map explains why its relatives, from Vyleesi® to KPV, do such different things.

KPV is the anti-inflammatory tail of alpha-MSH, studied mostly in cells and animals. It's not FDA-approved, and any use should go through a licensed provider.

## Frequently Asked Questions

### What does alpha-MSH stand for?

Alpha-MSH stands for alpha-melanocyte-stimulating hormone. It's named for its role in stimulating melanocytes, the skin cells that make pigment. It was later found to influence appetite, inflammation and sexual function too. It belongs to the melanocortin family, made from a precursor protein called POMC.

### What is the function of alpha-MSH?

Alpha-MSH has four main roles. It darkens skin pigment through MC1R, lowers appetite through brain MC4R, calms inflammatory signaling in immune cells, and influences sexual arousal. It does this by binding melanocortin receptors spread across the skin, brain, immune system and glands.

### Is KPV the same as alpha-MSH?

No. KPV is only the last three amino acids of alpha-MSH: lysine, proline and valine. It's studied for anti-inflammatory effects without alpha-MSH's pigment-darkening action. Most KPV research comes from cell and animal models. KPV is not FDA-approved, and use requires a provider's review.

### Does alpha-MSH suppress appetite?

Yes, in the brain. POMC neurons in the hypothalamus release alpha-MSH onto MC4R receptors, which lowers food intake and raises energy use. This pathway is why MC4R is a drug target. Setmelanotide, an MC4R agonist, is FDA-approved for weight management in certain rare genetic forms of obesity.

### Is Melanotan II the same as alpha-MSH?

No. Melanotan II is a synthetic, cyclic analog of alpha-MSH built to last longer in the body. It activates several melanocortin receptors, which is why it affects both tanning and arousal. Melanotan II is not FDA-approved, and products sold online often lack quality controls.

### What raises alpha-MSH naturally?

UV exposure raises alpha-MSH in the skin, which is part of tanning. Leptin, a hormone from fat tissue, switches on POMC neurons that release alpha-MSH in the brain. Stress also activates POMC processing. There's no proven diet or supplement that reliably raises alpha-MSH in humans.
